Hi saravanan,
	Pfaedit will be enough for this.
The most simple way is to open an existing malayalam Opentype font,
and the ttf font, and then copy each glyph from ttf font 
to curresponding place in opentype font. Delete any extra glyph
there in the opentype font.

Issue will be when you have any glyph in the ttf font which is 
not there in the opentype font.(Almost all common glyphs are 
available in the AkruthiMal2Normal font available from smc,
so this wont be a problem). Copy the glyph to end of the opentype
font, select its ligature (cntl+i), in the legature field 
type which all unicode chars are used to make this font...
